L5 Mutation Worksheet (Graded Assignment) This activity presents you with a small segment within a human gene that encodes for a protein and three point mutations that occur in the same set of three nucleotides in this gene segment. I recommend that you review the demo done in the L5 YouTube before tackling this assignment. 1) NORMAL (WILD TYPE) GENE SEGMENT The fourth set of three nucleotides, highlighted in yellow, is of interest. 3’ G A C T G A G G A C T C C T C T T C A G A a) Transcribe the entire gene segment b) Translate the mRNA to the corresponding polypeptide using the codon chart provided c) Describe the fourth amino acid in this gene segment in terms of its chemistry (i.e. is it charged? polar? nonpolar? hydrophobic? acidic? basic?) Recall that the R groups of amino acids contain functional groups that dictate the tertiary structure of the protein. 2) FOR EACH MUTANT: a) Just transcribe the mutated DNA sequence (this is fourth set of three nucleotides in the DNA, highlighted in yellow); b) Translate the mRNA to the corresponding amino acid using the codon chart i) What is the new amino acid? ii) Describe each new amino acid in terms of its chemistry. iii) How does each amino acid compare to the normal amino acid? iv) Finally, which point mutation proved to be the most detrimental to the tertiary structure of the original (wild type) protein?
